What is ATX-304?
O-304 (ATX-304) is a pan-AMPK activator developed by Betagenon in Sweden. It works by stopping AMPK from being switched off.
It’s studied in diabetes and heart function.
How ATX-304 works
AMPK is the cell’s energy sensor. When energy runs low it switches on glucose uptake and fat burning and switches off energy-hungry building processes.
O-304 doesn’t switch AMPK on directly. It stops phosphatases from removing the activating phosphate, so AMPK stays active longer once it’s triggered. In mice that improved glucose uptake in muscle and heart function.

Steneberg and colleagues reported in JCI Insight (2018) that O-304 lowered blood glucose and improved heart function in mice.
A small phase 2a trial in type 2 diabetes followed.
